1019 Cutibacterium acnes modulates the Pam3Csk4-induced innate immune responses in human keratinocytes

چکیده
Various insults can trigger innate immune activation through pattern recognition receptors(PRRs) in different cell types. These leave lasting epigenetic changes, and affected cells undergo cellular reprogramming, resulting altered responsiveness to subsequent encounters. This is an adaptive response of the system called memory(IIM). We were interested whether Cutibacterium acnes (C. acnes) may initiate similar events keratinocytes. used C. for primary training, after five days rest, Pam3Csk4 (TLR1/2 agonist) secondary induction normal human epidermal keratinocyte (NHEK) HaCaT cells. The mRNA expression several immune-related genes (e.g., TNFa, IL-8, SOCS1, TNFAIP3, TNIP1) increased Pam3Csk4-induced trained cells, compared Pam3Csk4-induced, untrained ones, suggestive training-like IIM processes. Expression differences also observed NHEK but their direction was not same originating from body parts. found statistically significantly higher TNFa levels breast region (NHEK-B), lower ones abdominal (NHEK-A) samples, indicative training vs., tolerance events, respectively. global 5-methylcytosine (5-mC) content genomic DNA isolated these samples control (untrained, uninduced) NHEK-B NHEK-A even 5 resting. Previous treatment led a marked sustained decrease cultures no changes ones. Members our microbiota modify that profoundly affects cutaneous responses vitro possibly vivo. Epigenetic representing skin regions arise due variations inhabiting composition.

Chitin Modulates Innate Immune Responses of Keratinocytes
BACKGROUND Chitin, after cellulose the second most abundant polysaccharide in nature, is an essential component of exoskeletons of crabs, shrimps and insects and protects these organisms from harsh conditions in their environment. Unexpectedly, chitin has been found to activate innate immune cells and to elicit murine airway inflammation. The skin represents the outer barrier of the human host ...
متن کاملCinobufagin Modulates Human Innate Immune Responses and Triggers Antibacterial Activity
The traditional Chinese medicine Chan-Su is widely used for treatment of cancer and cardiovascular diseases, but also as a remedy for infections such as furunculosis, tonsillitis and acute pharyngitis. The clinical use of Chan-Su suggests that it has anti-infective effects, however, the mechanism of action is incompletely understood. In particular, the effect on the human immune system is poorl...
متن کاملPRINS Non-Coding RNA Regulates Nucleic Acid-Induced Innate Immune Responses of Human Keratinocytes
Cytosolic DNA fragments are recognized as pathogen- and danger-associated molecular patterns that induce a cascade of innate immune responses. Moreover, excessive cytosolic DNA can enhance chronic inflammation, predominantly by activating inflammasomes, and thereby contributing to the pathogenesis of chronic diseases, such as psoriasis. Psoriasis associated non-protein coding RNA induced by str...
متن کاملLPLUNC1 modulates innate immune responses to Vibrio cholerae.
BACKGROUND Recent studies demonstrate that long palate, lung, and nasal epithelium clone 1 protein (LPLUNC1) is involved in immune responses to Vibrio cholerae, and that variations in the LPLUNC1 promoter influence susceptibility to severe cholera in humans. However, no functional role for LPLUNC1 has been identified. METHODS We investigated the role of LPLUNC1 in immune responses to V. chole...
متن کاملHuman Metapneumovirus Antagonism of Innate Immune Responses
Human metapneumovirus (hMPV) is a recently identified RNA virus belonging to the Paramyxoviridae family, which includes several major human and animal pathogens. Epidemiological studies indicate that hMPV is a significant human respiratory pathogen with worldwide distribution. It is associated with respiratory illnesses in children, adults, and immunocompromised patients, ranging from upper re...
